By playing games. And if you win, you get quite a bit of ip.

Okay, ip (influence points) are earned when you play games on League. But only when you do co-op and normals. Customs don't count unless it's a full team of 3v3 or 5v5. Ip is used to purchase champs in the game.

You can also buy Riot Points with money by using prepaid cards or using a credit card in the shop when you open the launcher. Riot Points can get things like skins and some summon icons and other things. You can mystery gift skins and champs to people by using RP, too.
